Global hemoglobin testing market by chromatography

Affinity chromatography, high-performance liquid chromatography (HPLC), and cation-exchange chromatography techniques are common technologies used to estimate blood hemoglobin and its variants such as hemoglobin F (HbF) or to check the concentration of HbA1c in people with diabetes. The HbF analysis is done in pregnant women in order to check the concentration of HbF in fetal blood to understand whether the fetus is a carrier of thalassemia or not.

In boron affinity chromatography, HbA1c binds with the boronate affinity resin, separating from the non-HbA1c. On further elution with sorbitol, HbA1c is released from the boronic acid sites on the resin, and its percentage is calculated from the photometric absorbance techniques.

In HPLC, HbA1c levels are determined by comparing the peak areas of the HbA1c and hemoglobin A in the chromatogram. Based on HPLC, ARKRAY, one of the prominent vendors in this market, develops ADAMS A1c HA-8180, which is an automated glycohemoglobin analyzer. Cation-exchange chromatography is used to find out the hemoglobin variants.

Global hemoglobin testing market by immunoassay

The interaction between antigen molecule (HbA1c) and specific antibody-coated latex beads (HbA1c specific) help to detect the concentration of HbA1c and total hemoglobin in the whole blood. This method is known as immunoassay method or latex-enhanced immunoassay method. The interaction results in the inhibition of agglutination. Inhibited agglutination decreases the scattering of light, resulting in reduced absorbance. A calibrated curve of absorbance and corresponding HbA1c concentration shows the HbA1c percentage from the absorbance readings. Siemens Healthcare developed DCA Vantage Analyzer, which is a POC immunoassay analyzer that helps in the quantification and detection of HbA1c in whole blood.

Global hemoglobin testing market by other techniques

The other segment includes capillary electrophoresis, direct enzymatic assay, and proteomic analysis by mass spectrometry. Capillary electrophoresis can be used to separate hemoglobin variants and hemoglobin derivatives. For instance, Sebia Electrophoresis developed CAPILLARYS 2 Neonat Fast capillary electrophoresis system, which works with the capillary electrophoresis technology to find variants in the blood of newborns. The micro-optical detection method utilizes immunoassays and chemistry technology to measure hemoglobin test results with the help of a handheld monitor and single-use test cartridges. “Mass spectrometry is used to detect masses in globin chain to identify a variant of hemoglobin. This kind of assay measures both HbA1c, variants of hemoglobin, and total hemoglobin levels,” says Barath.
